Application of the UHPLC method with PDA and emission detectors to study spectral and photophysical properties of 7-aminocoumarin, C120.

Abstract

Tematyka rozprawy doktorskiej skupia się na nietypowym zastosowaniu układu UHPLC z detektorem absorpcyjnym UV-VIS (PDA) i emisyjnym (FL) do badania właściwości spektralnych i fotofizycznych związków chemicznych, które mogą tworzyć kilka różnych indywiduów. W pracy zaproponowano koncepcję badań i opracowanie metodyki pomiarów, których celem jest wyznaczenie na układzie UHPLC-PDA-FL stałych homodimeryzacji o bardzo dużych wartościach, KDM = 105-1010 dm3/mol. Wyznaczenie tak dużych wartości KDM pozwoli po raz pierwszy na stwierdzenie w jakiej postaci występuje badany związek w szerokim zakresie stężeń 10-10 – 10-5 mol/dm3. Za pomocą układu UHPLC-PDA-FL możliwe jest, po rozdziale na kolumnie chromatograficznej, określenie jakie indywidua tworzy badany związek, wyznaczenie ich stężenia i zbadanie ich właściwości. Do badań wybrano 7-amino-4-metylo-1,2-benzopiron (C120). W wyniku przeprowadzonych badań i analizy wyników stwierdzono, że C120 w ACN tworzy szereg indywiduów: monomer, trzy kompleksy monomeru z cząsteczkami rozpuszczalnika, dwa dimery oraz dwa kompleksy dimerów z cząsteczkami rozpuszczalnika. Wyznaczono właściwości spektralne i fotofizyczne tych indywiduów. Stwierdzono również, że C120 w zakresie stężeń 10-6–10-4 mol/dm3 występuje niemal wyłącznie w postaci dimeru, a nie monomeru jak do tej pory powszechnie przyjmowano. Dla cząsteczki C120 wyznaczono bardzo dużą wartość KDM = 1.5 x 109 dm3/mol w ACN i w mniej polarnym 1-chlorobutanie, KDM = 9 x 109 dm3/mol.
The research work reported in the doctoral dissertation is concerned with an untypical use of the system UHPLC with an absorption detector UV-VIS (PDA) and emission detector (FL), for investigation of spectral and photophysical properties of chemical compounds that can form a few individua. A new conception of study was proposed and the appropriate methodology was devised for determination of modimerization constants of values from the range KDM = 105-1010 dm3/mol, using the system UHPLC-PDA-FL. Determination of so high homodimerization constants will permit for the first time for identification of the individua formed by a given compound in a wide range of concentrations 10-5 – 10-10 mol/dm3. The use of UHPLC-PDA-FL, after separation on a chromatographic column, permits identification of the individua made by a given compound and determination of their concentrations and properties. The measurements were carried out for 7-amino-4-methyl-1,2-benzopiron (C120). In acetonitrile C120 was found to form a number of individua, including monomer, three monomer complexes with the solvent, two dimers and two dimer complexes with the solvent. Spectral and photophysical properties of these individua were established. In the range of concentrations 10-6–10-4 mol/dm3, C120 was proved to occur almost exclusively in the form of dimer, not monomer as had been always assumed. For C120 the value of homodimerization constant was very high, in ACN it was KDM = 1.5 x 109 dm3/mol, while in less polar 1-chlorobutane, it was KDM = 9 x 109 dm3/mol.
