Geny integraz i oporności na antybiotyki β-laktamowe i glikopeptydowe rezystomu ścieków miejskich

Abstract

Występowanie u bakterii lekooporności jest niekwestionowanym problemem w leczeniu zakażeń i, wg WHO, jednym z trzech najpoważniejszych globalnych zagrożeń XXI wieku. Występowanie integronów i genów oporności na antybiotyki w środowisku uważane jest za zanieczyszczenie biotyczne i problem ekologiczny. Celem pracy była jakościowa i ilościowa analiza rodzaju i częstości występowania integronów klasy 1 oraz genów warunkujących oporność na antybiotyki β-laktamowe, karbapenemowe oraz glikopeptydowe w genomach bakterii hodowalnych, a także w metagenomie ścieków Centralnej Oczyszczalni Ścieków w Koziegłowach koło Poznania. Wykazano, że w trakcie oczyszczania ścieków następuje zwiększenie częstości występowania bakterii z genami stwarzającymi największe zagrożenie dla zdrowia publicznego, m.in. warunkującymi wytwarzanie β-laktamaz o rozszerzonym spektrum substratowym, karbapenemaz, a także wankomycynoopornych enterokoków i S. aureus opornych na meticylinę. Ponadto stwierdzono, że oczyszczanie ścieków prowadzi do zwiększenia częstości występowania integronów klasy 1 i genów oporności na β-laktamy i karbapenemy w rezystomie ścieku oczyszczonego, zrzucanego do rzeki Warty. Wyniki badań dostarczyły informacji na temat ograniczonej skuteczności oczyszczania ścieków w eliminacji bakterii antybiotykoopornych i genów oporności, wskazując tym samym na potrzebę opracowania efektywnej strategii i wdrożenia dodatkowych metod dezynfekcji ścieków, w celu ograniczenia narastania i rozprzestrzenianie się oporności na antybiotyki w środowisku.

The occurrence of drug resistance in bacteria is an unquestionable problem in the treatment of infections and, according to the WHO, one of the three most serious global threats in the 21st century. The presence of integrons and antibiotic resistance genes in the environment is considered biotic pollution and ecological problem. The aim of this study was the qualitative and quantitative analysis of type and frequency of class 1 integrons and genes conferring resistance to β-lactam, carbapenem and glycopeptide in the genomes of culturable bacteria, as well as in the wastewater metagenome of Central Wastewater Treatment Plant in Koziegłowy near Poznań. The results indicate that during the wastewater treatment there is an increase of the frequency of bacteria with genes that pose the greatest threat to public health, including genes of extended spectrum β-lactamases, carbapenemases, as well as enterococci resistance to vancomycin and S. aureus resistance to methicillin. Moreover, it was found that the wastewater treatment process leads to increased frequency of class 1 integrons and resistance genes to β-lactam and carbapenem in resistome of final effluent, discharged to Warta River. Results of this study provided significant information on the limited effectiveness of wastewater treatment processes in the elimination of antibiotic-resistant bacteria and resistance genes, which indicates the need to develop an effective strategy and implement additional methods of wastewater disinfection, in order to limit the increase and spread of antibiotic resistance in the environment.
