Synthesis and structure of new amino analogues of ansamycin antibiotic rifampicin

Abstract

Na drodze aminowania redukcyjnego, różnymi metodami, zsyntezowano 18 (AR1-AR18) nowych aminowych analogów ansamycynowego antybiotyku rifampicyny (Rif). Ustalono, że najlepszą metodą syntezy aminowych analogów AR1-AR18 jest aminowanie redukcyjne z użyciem NaBH3CN w obecności HCl, jako katalizatora. Na podstawie danych uzyskanych z widm 1D NMR: 1H, 13C, 15N i 2D NMR: 1H-1H COSY, 1H-13C HSQC, 1H-13C HMBC, 1H-1H NOESY, 1H-15N HSQC, 1H-15N HMBC oraz FT-IR ustalona została struktura AR1-AR18 i Rif w rozpuszczalnikach aprotycznych i protycznych. Analiza danych spektroskopowych z wynikami badań krystalograficznych wykazały, że niezależnie od rodzaju użytego rozpuszczalnika AR1-AR18 występują w formie zwitterjonowej, podczas gdy dla Rif wewnątrzcząsteczkowe przeniesienie protonu jest realizowane w rozpuszczalnikach protycznych. Wyniki testów aktywności biologicznych Rif i AR1–AR18 w połączeniu z analizą oddziaływań ligand-enzym wskazały, że za ekstremalnie wysoka aktywność przeciwbakteryjna Rif jest zależna od miejsca protonacji. Zaproponowano nowy model inhibicji bakteryjnej polimerazy RNA dla ansamakrolidów. Na podstawie danych uzyskanych z badań tandemowych MALDI-TOF MS w trybie jonów dodatnich zaproponowano drogi fragmentacji masowej Rif oraz AR1–AR7. Biorąc pod uwagę wszystkie dane spektrometryczne dowiedziono, że fragmentacja Rif i jej AR1–AR7 analogów jest głównie przeprowadzana na moście ansamycynowym. Na drogi rozpadu masowego nowych aminowych analogów AR1–AR7 nie ma wpływu natura i wielkość podstawnika przy atomie węgla C(38). Sygnały od jonów B, K i N ze względu na ich relatywnie wysoką intensywność oraz obecność we wszystkich widmach analogów mogą służyć do wykrywania tego typu związków np. w materiałach biologicznych.
Eighteen AR1-AR18 new amino analogues of ansamycin antibiotic rifampicin (Rif) have been synthesized via different reductive amination methods. It was indicated that the most efficient method of amino analogues` synthesis is reductive alkylation with the use of NaBH3CN in the presence of HCl as catalyst. On the basis of 1D NMR: 1H, 13C, 15N and 2D NMR: 1H-1H COSY, 1H-13C HSQC, 1H-13C HMBC, 1H-1H NOESY, 1H-15N HSQC, 1H-15N HMBC as well as FT-IR data the structure of AR1-AR18 and Rif in protic and aprotic solvents has been determined. NMR analysis together with the X-ray data evidenced intramolecular proton transfer in rifampicin and its analogues AR1–AR18 with the formation of zwitterions, irrespectively of the kind of the solvent. In contrast, for Rif the intamolecular proton transfer have been indicated only in protic solvents with addition of water. Biological tests of Rif, AR1–AR18 in combination with the analysis of ligand–protein interactions have revealed the relationship between the protonation site and extremely high antibacterial activity and led to presentation of new model of inhibition of bacterial RNA polymerase via this class of ansa macrolides. The fragmentation pathways of Rif and its seven new aza-alkyl analogues (AR1–AR7) were investigated by MALDI-TOF tandem MS in positive ion detection mode Taking all the mass spectrometric data into account, it can be concluded that the mass decompositions of Rif and its AR1–AR7 analogues under MALDI method are mainly realised by fragmentation of ansa bridge of antibiotics. As a result of fragmentation of ansa bridge different product ions are formed because of the presence of Rif and AR1–AR7 structures’ different functional groups: hydrazone for Rif and amine for AR1–AR7. The fragmentation pathways of novel aza-analogues of rifampicin are independent of the nature and the size of the substituent at C(38) atom. Ions B, K and N signal because of their relatively high abundances and their compositions (all contain not fragmented amine substituent part at C38) are suitable for analytical detection and identification of this type of rifampicin analogues.
