dc.description.abstractAntybiotyki β-laktamowe z grupy penicylin należą do najważniejszych leków stosowanych w terapii zakażeń bakteryjnych. Ze względu na obecność licznych grup funkcyjnych mogą tworzyć związki kompleksowe z jonami metali, co może wpływać na ich właściwości fizykochemiczne, trwałość, biodostępność i aktywność biologiczną. Celem rozprawy było określenie wpływu podstawników antybiotyków β-laktamowych z grupy penicylin na ich zdolność do kompleksowania z jonami metali bloku s, d i f o znaczeniu biologicznym bądź diagnostycznym. Badania przeprowadzono z wykorzystaniem miareczkowania potencjometrycznego połączonego z komputerową analizą danych oraz metod spektroskopowych. Wyznaczono skład, ich termodynamiczne stałe trwałości (logβ) i stałe równowagi reakcji tworzenia (logKe), a także określono miejsca koordynacji jonów metali i budowę wewnętrznej sfery koordynacyjnej. Analizy potwierdziły proces kompleksowania oraz umożliwiły identyfikację grup funkcyjnych uczestniczących w koordynacji jonów metali. Uzyskane wyniki wykazały, że charakter podstawników w cząsteczkach penicylin istotnie wpływa na ich właściwości kompleksujące. Przeprowadzone badania poszerzają wiedzę na temat właściwości koordynacyjnych antybiotyków β-laktamowych oraz ich związków kompleksowych. β-Lactam antibiotics from the penicillin group are among the most important drugs used in the treatment of bacterial infections. Due to the presence of numerous functional groups, they can form complex compounds with metal ions, which may affect their physicochemical properties, stability, bioavailability, and biological activity. The aim of this dissertation was to determine the influence of substitutes present in β-Lactam antibiotics from the penicillin group on their ability to form coordination compounds toward selected biologically or diagnostically relevant s-, d-, and f-block metal ions. The study combined potentiometric titration with computer-assisted data analysis and spectroscopic techniques. The composition of the complexes, their overall stability constants (logβ), and equilibrium constants of formation (logKe) were determined. In addition, the coordination sites of the metal ions and the structure of the inner coordination sphere were identified. Spectroscopic analyses confirmed the complexation process and enabled the identification of the functional groups involved in metal ion coordination. The obtained results demonstrated that the nature of the substituents in penicillin molecules significantly influences their complexing properties. The findings expand current knowledge of the coordination properties of β-lactam antibiotics and their metal complexes.
